Hello,
I have configured a broker, a thing with channels and the items using configuration files ( without Paper UI).
This all is working - but only with the manual link (from the channel to the item) in the Paper UI - what is wrong ?

Welcome to the openHAB forum 
You ask what is wrong? That is “easy” to tell, the used syntax in your setup files which should create the link is wrong ( since “everything” else is working). So, use the correct syntax and you are set!
